The third-placed Whites beat league leaders Leicester last week and have closed the gap to second-placed Ipswich to eight points with four wins in five.
Argyle, who are 19th, are yet to win away from home this campaign, while Leeds are unbeaten at Elland Road.
“We have to be more spot on for this game than we were for the Leicester match,” he said.
“For me, two situations in the season are really important. One is when you have a setback, how you react. We have had this already this season and we have reacted in a good manner with performances and results.
“The other is when you win the highlight game and everyone applauds you, and the players and supporters are having a great time celebrating their incredible victory… You need to make sure that you are even more focused on winning the following game, when everyone is expecting you to win.
“You cannot enter a game in this league thinking it’s just another workday.
It’s going to be a season-defining game for Plymouth. They have put a lot of effort into organizing an away game at Elland Road, and if we approach it as just another game, we won’t have a chance to gain the points.
Pascal Struijk, a defender who is always there for Leeds, will be missed because he requires hernia surgery.
Farke has been impressed by the League One champions from the previous season, despite the fact that the Pilgrims had only won one of their last seven games.
“They’re a team that is used to winning football games from last season and you can see they still play with that confidence and freedom,” he stated on BBC Radio Leeds.
“They were the better team for a much of the game at Ipswich and were unfortunate to lose. That says a lot about how good they are.
